BASIC FUNCTION AND RESPONSIBILITY
Perform laboratory maintenance and preparation of stock solutions. Perform experiments using behavioral, physiological, biochemical, cell and molecular methodologies in mice, mouse brain tissue and primary neuronal cultures. Keep detailed written and organized records of experimental design, protocols, computer data analysis and raw data. Maintain records of laboratory reagents, supplies, freezer storage logs, plasmids and oligonucleotide inventories.
CHARACTERISTIC DUTIES AND RESPONSIBILITIES
Maintain colonies of genetically modified mice: breeding, weaning and genotyping of the mice as well as maintaining electronic records of the colonies.
Perform experiments with mice. This includes stereotaxic injection of viral vectors into the specific region of mouse brain, and fiber optic cannula implantation for performing optogenetic and chemogenetic experiments. It also includes comprehensive behavioral testing in transgenic mice mimicking human genetic risk of eating disorders and obesity, and a number of cell biological, physiological, and biochemical experiments investigating the function of brain circuits. Perform sterile techniques in mice and a cell culture facility for the preparation and maintenance of primary neuronal cultures from genetically modified mice.
Perform cell and molecular biology experiments using mouse tissues. These include light and electron microscopic analyses of protein and organelle localization, preparation of mRNA, analysis of gene expression by quantitative PCR, preparation of native and denatured protein extracts, protein assays, analysis and quantitation of proteins and phosphoproteins using Western blots.
Operate and organize the maintenance of a variety of laboratory instruments including pH meters, centrifuges, cell culture hoods, microscopes, quantitative PCR instruments, thermal cyclers, and image acquisition and analysis instruments. Prepare laboratory reagents, e.g. buffers and cell culture media.
REQUIRED QUALIFICATIONS
A Bachelor's degree in psychology and biology or an equivalent combination of education and progressively responsible experience in a neuroscience or molecular biology related field.
Excellent written and verbal communication skills as well as interpersonal relationship skills.
Experience with a variety of emotional and reward behavioral assays in mice.
Experience with recent advanced modern neuroscience techniques including chemogenetics, optogenetics and electrophysiology
Experience with stereotaxic microinjection of viral vectors into mouse brain.
Experience with brain histological and histochemical techniques, including transcardial perfusion, brain sectioning, and immunofluorescence staining.
Experience operating epifluorescence, confocal, and electron microscopes.
Experience with handling, breeding, and genotyping mice.
Experience with data analysis (including basic statistics) and presentation using Excel, GraphPad Prism.
DESIRABLE QUALIFICATIONS
Experience with basic cell and molecular biology including but not limited to the preparation of protein extracts, RNA preparation, quantitative PCR and Western blots.
Experience in the quantitative analysis of gene and protein expression.
Experience in the quantitative analysis of EEG/EMG data for sleep architecture analysis.
RATE OF PAY
$21.15 hourly rate
